چکیده

Impairment of the insulin regulation of energy metabolism is considered to be an etiologic key component for metabolic disturbances. Methods for studies of insulin sensitivity thus are highly topical. There are clear indications that reduced insulin sensitivity contributes to the metabolic disturbances that occurs especially among obese lactating cows. Direct measurements of insulin sensitivity are laborious and not suitable for epidemiological studies. We have therefore adopted an indirect method originally developed for humans to estimate insulin sensitivity in dairy cows. The method, "Revised Quantitative Insulin Sensitivity Check Index" (RQUICKI) is based on plasma concentrations of glucose, insulin and free fatty acids (FFA) and it generates good and linear correlations with different estimates of insulin sensitivity in human populations. We hypothesized that the RQUICKI method could be used as an index of insulin function in lactating dairy cows. We calculated RQUICKI in 237 apparently healthy dairy cows from 20 commercial herds. All cows included were in their first 15 weeks of lactation. RQUICKI was not affected by the homeorhetic adaptations in energy metabolism that occurred during the first 15 weeks of lactation. In a cohort of 24 experimental cows fed in order to obtain different body condition at parturition RQUICKI was lower in early lactation in cows with a high body condition score suggesting disturbed insulin function in obese cows. The results indicate that RQUICKI might be used to identify lactating cows with disturbed insulin function.

Plasma pharmacokinetics of pioglitazone following oral or intravenous administration in Holstein cows

Pioglitazone belongs to the thiazolidinedione (TZD) class of antidiabetic agents, with proven efficacy in increasing insulin sensitivity and in the treatment of type 2 diabetes mellitus in humans. Pioglitazone has been proposed as a potential feed additive to reduce insulin resistance and consequently some of the metabolic disorders in transition cows. This study was aimed at determining the ph...

Relationship between Insulin to Glucagon Ratio and Metabolic Parameters in Primiparous and Multiparous Dairy Cows in Transitional Period

In order to investigate the relationship between insulin to glucagon ratio and metabolic factors in transitional period of dairy cows, 28 cows heifers with body condition score of 3.25- 3.75 were selected. Dairy cows received close up diet, from 21 days prepartum until parturition and lactation diet according to nutritional requirements after parturition. Blood was sampled at 10 days before and...

P-64: The Relationship between Polymorphism in Gene of Insulin-Like Growth Factor-I and The Serum Periparturient Concentration in Holstein Dairy Cows

Background: One of the most important metabolic factors affecting the reproductive activity is insulin-like growth factor-I (IGFI) concentration changes after calving. Recently, Maj et al. (2008) discovered a significant association between the IGF-I genotypes based on the 5'-untranslatedregion (5'-UTR) of IGF-I gene and the IGF-I blood level. The objective of this study is to investi...

Metabolic profiles of high-yielding dairy cows with ovarian cysts formation

Background: Ovarian cysts are among the diseases which cause reproductive failure and economic losses in dairy herds. High yelding dairy cattles are suseptable to reproductive failure caused by ovarian cysts, as a result of their exposure to stressful coditions during lactation. OBJECTIVES: This study aimed to monitor metabolic profile tests in dairy cows with ovarian cysts in comparison with c...

Comparison of Various Indices of Energy Metabolism in Recumbent and Healthy Dairy Cows

BACKGROUND Downer cow syndrome (DCS) is often diagnosed in dairy cattle during the early post-partum period. The etiology of this condition is not completely understood, as it can be related to the energetic or electrolyte metabolism, as well as to infectious diseases or to trauma. HYPOTHESIS/OBJECTIVES The aim of this study is to compare energy metabolism and insulin sensitivity indices and ...
